Engineered therapeutic nanovaccine against chronic hepatitis B virus infection

Chronic hepatitis B (CHB), caused by persistent virus (HBV) infection, significantly increases the risk of leading to liver diseases. Despite successful development and implementation HBV prophylactic vaccines for several decades, therapeutic vaccine, a substantially potential strategy eradicate achieve CHB cure, remains great challenge. Herein, we applied flash nanocomplexation (FNC) technology prepare nanovaccines with narrow size distribution high encapsulation via charge complexation between chitosan heparin encapsulate recombinant surface antigen (rHBsAg) or core (rHBcAg), CpG as adjuvant. The two enhanced uptake adjuvant into Raw264.7 cells their co-administration further promoted maturation activation bone marrow-derived dendritic (BMDCs). Meanwhile, they exhibited excellent lymph nodes (LNs) targeting ability, draining proximal distal LNs prolonged retention time, following subcutaneous injection. Co-administered could break immune tolerance restore HBV-specific responses. In mouse model CHB, 90% 80% mice achieved (HBsAg) seroclearance antibody (HBsAb) seroconversion, respectively. Moreover, induced long-term memory in HBV-cured protect them from reinfection. Thus, this work offers promising translational alternative vaccine.
